🧬 Family & Essentiality

Vitamin FamilyFat-soluble vitamin
EssentialityEssential
FormsTocopherols & Tocotrienols (α, β, γ, δ)

🌿 Plain-Language Summary

Vitamin E is a family of fat-soluble antioxidants that help protect polyunsaturated fats in cell membranes from oxidative stress. The most biologically active form in humans is α-tocopherol, used as the reference for Nutrient Reference Values (NRVs).

⚗️ Forms & Molecular IDs

  • Tocopherols: α, β, γ, δ (saturated phytyl side chain)
  • Tocotrienols: α, β, γ, δ (unsaturated isoprenoid side chain)
  • Natural: RRR-α-tocopherol
  • Synthetic: all-rac-α-tocopherol

🆚 Tocopherols vs Tocotrienols

Side Chain: Tocopherols have saturated tails (rigid); Tocotrienols have unsaturated tails (more flexible, better bilayer penetration).
Biological Retention: α-Tocopherol is selectively maintained in plasma by α-TTP. Tocotrienols are less well retained.
Dietary Sources: Tocopherols in nuts, seeds, and vegetable oils; Tocotrienols in palm oil, rice bran, barley, annatto.
Functions: Both act as antioxidants. Tocopherols dominate human tissues. Tocotrienols show emerging roles (cholesterol regulation, neuroprotection, anti-inflammatory actions) but evidence is still evolving.

⚡ Functions

  • Protects membrane polyunsaturated fatty acids (PUFAs) from oxidation
  • Works synergistically with vitamin C, selenium (via GPx enzymes), and CoQ10
  • Supports immune defence and red blood cell stability

🍽️ Absorption & Bioavailability

May increaseDietary fat, mixed meals, bile & pancreatic sufficiency
May decreaseFat-malabsorption, orlistat, bile acid sequestrants, very low-fat diets

Requirements are expressed as mg α-TE (α-tocopherol equivalents).

🥗 Food Sources

  • Plant oils: sunflower, safflower, wheat germ, canola, olive
  • Nuts & seeds
  • Avocado; wholegrains
  • Tocotrienol-rich sources: palm oil, rice bran, annatto, barley

🇦🇺 Australian NRVs (NHMRC)

  • Adults (AI): Men 10 mg/day; Women 7 mg/day (as mg α-TE)
  • Pregnancy: 7 mg/day
  • Lactation: 11 mg/day
  • UL (adults): 300 mg/day (as mg α-TE)

🚨 Deficiency & Excess

Deficiency (rare)

Peripheral neuropathy, ataxia, and muscle weakness — usually in fat-malabsorption states or rare genetic disorders.

Excess

Very high doses may increase bleeding tendency.

🧪 Testing & Monitoring

  • Serum/plasma α-tocopherol measurement
  • Sometimes expressed relative to lipids (e.g., α-tocopherol:cholesterol ratio)

📊 Evidence Snapshot

  • 🔬✅ Well-established: Lipid-phase antioxidant; deficiency syndromes in fat-malabsorption
  • 🧪⚖️ Promising/mixed: Tocotrienols show potential roles in neuroprotection & cholesterol modulation, but evidence is evolving
  • ⚠️❌ Unproven/hype: High-dose supplementation for broad anti-ageing or performance enhancement
